In a translational fusion, the reporter gene (e.g. lacZ, GFP) is fused in the same reading frame as the target gene, producing a single chimeric fusion protein. This allows detection of both transcriptional and post-transcriptional regulation and can reveal protein localisation within cells. In contrast, a transcriptional fusion places the reporter downstream with its own RBS, detecting only promoter activity.
