S. cerevisiae is a eukaryote and can perform post-translational modifications (PTMs) including N-linked and O-linked glycosylation, signal peptide cleavage, and disulfide bond formation — processes absent in E. coli. Many therapeutic proteins (e.g. insulin, hepatitis B vaccine antigen) require these modifications for correct folding and biological activity, making yeast a preferred eukaryotic expression system.
