A metabolic bottleneck is an enzymatic step whose low activity or tight regulation limits the flux of carbon through the pathway toward the desired product. Removing a bottleneck (by overexpressing the enzyme gene, replacing it with a deregulated variant, or deleting competing pathways) can dramatically increase product yield. However, removing one bottleneck usually reveals the next rate-limiting step — hence, entire pathways often need 'engineering' for significant improvements.
