DNA polymerase is the enzyme responsible for synthesising new DNA strands by adding nucleotides complementary to a template strand (5′→3′ direction). It requires a primer and cannot initiate new chains de novo. DNA ligase joins Okazaki fragments; DNA topoisomerase relieves supercoiling ahead of the replication fork; RNA polymerase synthesises RNA, not DNA (except reverse transcriptase, which uses RNA as template).
