BRCA2 mutations are the most significant hereditary risk factor for male breast cancer. Men with BRCA2 mutations have a lifetime breast cancer risk of ~6–8% (vs ~0.1% in the general male population). BRCA2 mutations also increase risk of prostate and pancreatic cancer. While BRCA1 mutations also increase male breast cancer risk, the association is weaker. BRCA2 mutations account for the majority of hereditary male breast cancer cases (vs BRCA1, which predominantly affects female breast/ovarian cancer).
