Non-competitive inhibition can be either reversible or irreversible. The inhibitor binds to an allosteric site (not the active site), causing a conformational change that reduces enzyme activity. Unlike competitive inhibition, it cannot be overcome by increasing substrate concentration because the inhibitor does not compete with the substrate for the active site.
